"Cell damage caused by free radicals plays an important role in the pathogenesis of many animal and human diseases. The main aim of the project is to clarify the role of the nitrite oxide (NO) in epithelial cell damage caused by oxygen free radicals in consequence of oxidative stress. For this purpose appropriate models of epithelial cell lines damage by oxygen free radical donors will be proposed. Physiological changes and changes in intracellular redox state of epithelial cells will be examined afteroxidative stress and after treatment of cell by different concentrations of NO. Arachidonic acid and cyclic guanosin monophosphate signal transduction pathways in effect of NO on epithelial cells in consequence with oxidative stress will be studied with use of specific inhibitors of these signal pathways. Low molecular weight antioxidants N-acetylcysteine and reduced glutathione will be used to clarify mutual effect of NO and oxygen free radicals on epithelial cells.
