"The structural remodelling of peripheral pulmonary arteries is the main cause of the pulmonary hypertension developing during chronic hypoxia. The presence of small collagen cleavage fragments in pulmonary vessels suggests involvement of the matrixmetalloproteinases (MMPs). The MMPs may come from activated mast cells. The aim of this work is %22in vitro%22 study of the MMPs production in isolated rat pulmonary mast cells. We would like to prove that hypoxia stimulates production and release of MMPs bymast cells and detect MMPs subtypes including their proenzymes. The specific monoclonal anti-rat MMP-13 will be used for histochemical detection of MMP-13 in mast cells granulas. The other types of MMPs will be analysed by zymography. We will culture andactivate the mast cells on collagen type I to compare collagen cleavage fragments produced with fragments found in pulmonary arteries of chronically hypoxic rats.
