"The aim of the project is to enhance knowledge on the binding domains and functional properties of muscarinic receptors for acetylcholine by an examination of %22non-standard%22 interactions between mucarinic receptors and xanomeline (a substance with agonistic effects) and methoctramine (a substance with antagonistic effects). There are reasons to suppose that xanomeline associates with the receptors in the classical binding domain but also in another domain, with which it remains very firmly linked and from which it can act on the classical binding domain. In the case of methoctramine, experimental results suggest that it interacts with the classical binding domain and also (by the opposite end of its molecule) with another binding domain which hasnot yet been identified. We plan to perform genetic modifications of muscarinic receptors and to evaluate their effects on the interactions between the receptors and xanomeline and methoctramine.
