"Protein p73 vykazuje vysokou homologii s\u00A0bl\u00EDzce p\u0159\u00EDbuzn\u00FDm proteinem p53 a vyskytuje v\u00A0mnoha isoform\u00E1ch, kter\u00E9 se od sebe li\u0161\u00ED jak ve struktu\u0159e, tak funkci. Zjistilo se, \u017Ee p73 nen\u00ED klasick\u00FD n\u00E1dorov\u00FD supresor, ale je nezbytn\u00FD pro spr\u00E1vnou funkci proteinu p53 v\u00A0apoptotick\u00FDch drah\u00E1ch a tak\u00E9 hraje (spolu s\u00A0p63) v\u00FDznamnou roli ve v\u00FDvoji organismu. It exists in many isoforms that differ in their structure and functions. It was found that p73 is not a classic tumour suppressor, but it is crucial for proper function of p53in apoptotic pathways and also plays (together with p63) an important role in development of an organism. The p73 proteins are able to induce p53-target genes (such as p21, mdm2, bax, etc.). This project is focused on studies of the p73 DNA binding properties of protein p73 in vitro, in comparison with the properties of the p53 protein. We will investigate p73 interactions with different target sites in supercoiled or linear DNA. Next we will study the influence of formation of %22non-B%22 DNA structures stabilised by the DNA supercoiling (such as cruciforms) within the target sites, as well as influence of DNA modification with antitumor agents, on the p73-DNA binding.
