"Na+/H+-antiporters are secondary active transport systems occurring in almost all types of cells. The family of homologous yeast plasma membrane Na+/H+-antiporters includes several genes from four different yeast species and can be divided in twodistinctsubfamilies: (1) antiporters with substrate specificity only for Na+ and Li+ (Schizosaccharomyces pombe, Zygosaccharomyces rouxii) with primary detoxification function in cells and (2) antiporters (Saccharomyces cerevisiae, Candida albicans)mediating th e transport of at least four alkali metal cations (Na+, Li+, K+, Rb+) with multiple functions in cells. The main aim of this project is to identify essential amino acid residues or regions in the transmembrane part of yeast plasma membraneNa+/H+ antiporters determinating the substrate specificity for K+ (Rb+) transport. These regions will be identified (i) by mutagenesis of Z. rouxii Na+/H+-antiporter ZrSod2-22p (transports only Na+ and Li+) that would change and/or broaden the substrate"@en . . . . . . . .
