"Importance of the interaction of myeloperoxidase with the endothelial glycocalyx for leukocyte recruitment and vascular fuction"@en . . . . "1"^^ . . . . . "Deferred Final Report"@en . . "Activation and recruitment of polymorphonuclear neutrophils (PMN) is not only central to the innate immune response but also in the vascular inflammatory processes. Myeloperoxidase (MPO), expressed in large amounts in PMN and released upon their activation, evokes these processes in a decisive manner. Currently, with our collaborators we have shown that MPO induces motility of PMN that is dependent on electrostatic interactions. The interaction of highly cationic MPO with endothelial glycocalyx glycosaminoglycans would reduce the negative charge of glycocalyx that can detrimentally affect endothelium functions. The objective is to characterize to which extent MPO modulates the charge of the glycocalyx, the glycocalyx three dimensional structure and composition, and ultimately modulates endothelium permeability, recruitment of leukocytes, microcirculatory rheology and transduction of shear stress. Findings would significantly extend the knowledge of MPO inflammatory properties and identify previously unrecognized features of MPO as a potential pharmacological target."@en . "4"^^ . "0"^^ .
