"Our previous studies\u00A0have shown that endothelin system plays an important role in the development of hypertension and end-organ damage in homozygous Ren-2 rats and that interventions applied at youth are more efficient than when adopted in adulthood. It is known that animal models with exogenously administered angiotensin II (ANG II) exhibit an ET-dependent component, whereas hypertensive models with endogenously enhanced production of ANG II, do not. In addition, it has been shown that ETA receptors are responsible for end-organ damage, since their blockade significantly ameliorated these changes and moreover, had substantial impact on blood pressure (BP). ETB receptors seem to serve as opponents to ETA receptors mediating peripheral vasodilation and renal natriuresis. Their activation could therefore add to lowering of blood pressure. Ren-2 transgenic rats [TGR(mRen-2)27] together with a strain with inducible hypertension [Cyp1a1-Ren-2) are two models with ANG II-dependent hypertension enabling an insight into the mechanism of interaction between renin-angiotensin and endothelin systems.
