Inflammation is a prominent factor in cancer etiology; however, little is known about its impact on toxicity of carcinogenic PAHs. We propose to study the mechanisms, through which inflammatory mediators modulate specific toxic activities of PAHs. Using cells derived from respective target tissues, we will analyze the interactive effects of inflammatory mediators and PAHs (and their environmental mixtures) on the processes linked to carcinogenesis such as the metabolic activation of PAHs, DNA damage, disruption of cell proliferation/apoptosis control and altered cell-cell communication, which are related to activation of the aryl hydrocarbon receptor (AhR). We expect to obtain a principal novel information about interactions of PAHs and inflammatory mediators, which would significantly modulate the risks associated with exposure to PAHs, especially with regards to their genotoxicity and the effects related to tumor promotion."@en .
