"5"^^ . "The aim of the proposed project is to define signaling pathways and genes activated in cumulus-oocyte complexes (COCs) during resumption of meiosis in vitro. The resumption of meiosis is induced in vivo by a preovulatory surge of gonadotropin hormones \u2013 LH and FSH; the same stimulation is routinely used to trigger maturation of COCs in vitro. The gonadotropins induce synthesis of EGF-like peptides in cumulus cells surrounding the oocyte and initiate a dramatic change in expression of genes controlling resumption of oocyte meiosis, expansion of cumulus cells and steroidogenesis. We will find out what signaling pathways and genes are activated by the action of FSH/LH and EGF-like peptides in pig cumulus cells and what is their relation to oocyte meiotic maturation and expansion of cumulus cells. To achieve our goals, we will use progressive molecular genetics and biochemical methods, first of all immunoblotting, detection of signaling molecules by life-imaging, and transcriptome analysis of cumulus cells by microarray hybridization."@en .
