"Senescence has been defined as the endogenously controlled degradation process, which could be induced prematurely by different kind of stress. NO has been suggested to have a protective role against senescence but less information is available on NO-derived molecule \u2013 nitrogen reactive species (RNS). Hence, it is of special interest to determine whether the course of changes in nitrosative stress and protection against it are similar to the changes in oxidative stress and changes in antioxidant capacity. We will use transgenic tobacco with autoregulated senescence-induced production of cytokinins. These plants have increased life span and the changes of antioxidant protection during their senescence had been extensively studied. We will provide the first summary study of RNS during natural and during senescence induced by high levels of Zn with regard to the metabolism of nitrogen in quality of macronutrient. We will also study the application and modification of the Comet assay in plant senescence research and we will compare DNA damage (single and double strand breaks, alkali labile sites, incomplete excision repair sites and DNA cross-links) during both natural and stress induced senescence by the Comet or Single cell gel electrophoresis assay."@en .
