"Research on the single mitochondrion (mt) of the disease-causing parasite Trypanosoma brucei has revealed several biological oddities such as an expansive DNA network, byzantine RNA metabolism famously involving RNA editing. The organelle switches from a fully functional state in the insect-dwelling stage to a greatly reduced one in the pathogenic stage infecting the mammalian host. Several problems such as how the manifold steps required for expression of mt genes are spatially and temporally coordinated plus what is the shape of the mitoproteome between the two major life stages have yet to be solved. To address these and other questions, we propose to fluorescently tag up to 200 mt proteins to determine their sub-organellar and potential differential interstagial localization using the cutting edge techniques of recombinogenic engineering and super resolution microscopy. This project also aims to establish this platform for addressing the bigger question of the function of more than half of unknown proteins that comprise the whole T. brucei proteome."@en .
