Our intention is to characterize the function of all components of this pathway in the parasitic protist Trypanosoma brucei that can be identified by homology searches. Moreover, we also aim to identify novel components by operon and expression analyses. Initial studies indicate that there are significant differences in the Fe-S assembly between the procyclic (in tse-tse vector) and bloodstream stages (in mammalian blood), both in terms of protein components and localization within the cell. Functions of numerous Fe-S cluster-containing proteins (at wild type levels or overexpressed) will be used to monitor participation of individual proteins on the assembly.
