"The role of the Wnt signaling pathway in development and proper homeostasis of rapidly self-renewing epithelia of gastrointestinal tract is well established.Moreover, mutations in genes encoding components of Wnt signaling are directly linked to the onset of intestinal cancer. The first part of the project is focused on the Wnt signaling target gene Troy. The Troy gene encodes a single-pass transmembrane protein, a member of the tumor necrosis factor receptor superfamily, which is almost exclusively expressed in intestinal neoplasia. We will elucidate, using mouse strains prone to intestinal cancer,whether Troy-dependent signaling contributes to the tumor initiation and progression. The second part of the project is dedicated to the analysis of the phenotypic changes following conditional inactivation of the transcriptionfactors Tcf3 and Tcf4 in mouse intestine. Distinct sets of genes regulated by these nuclear effectors of the Wnt signaling pathway will be identified.
