Na\u0161e p\u0159edchoz\u00ED pr\u00E1ce ukazuj\u00ED, \u017Ee prenat\u00E1ln\u00ED expozice metamfetaminu (MA) m\u00E1 negativn\u00ED vliv na postnat\u00E1ln\u00ED v\u00FDvoj ml\u00E1\u010Fat laboratorn\u00EDho potkana, na je kognitivn\u00ED funkce v dosp\u011Blosti, ovliv\u0148uje nocicep\u010Dn\u00ED citlivost a n\u00E1chylnost ke vzniku epileptick\u00FDch z\u00E1chvat\u016F. \u00A0Na\u0161e nejnov\u011Bj\u0161\u00ED studie za pou\u017Eit\u00ED mikrodialyza\u010Dn\u00EDch metod ukazuje, \u017Ee prenat\u00E1ln\u00ED expozice MA m\u00E1 dlouhodob\u00FD \u00FA\u010Dinek na motoriku a metabolismus dopaminu dosp\u011Bl\u00FDch zv\u00ED\u0159at. Zjistili jsme, \u017Ee prenat\u00E1ln\u00ED expozice MA zvy\u0161uje hladinu dopaminu a jeho metabolit\u016F v nucleus accumbens (NAc) po n\u00EDzk\u00E9 stimula\u010Dn\u00ED d\u00E1vce MA (1 mg/kg) v dosp\u011Blosti. Existuje p\u0159edpoklad, \u017Ee v\u00A0\u010Dase mezi prvn\u00EDm oku\u0161en\u00EDm drogy a n\u00E1vykov\u00FDm (kompulzivn\u00EDm) chov\u00E1n\u00EDm jedince doch\u00E1z\u00ED v\u00A0mozkov\u00FDch struktur\u00E1ch k\u00A0posunu aktivace od prefront\u00E1ln\u00ED mozkov\u00E9 k\u016Fry (PFC) ke striatu a tam od jeho ventr\u00E1ln\u00ED \u010D\u00E1sti (NAc) k\u00A0dorz\u00E1ln\u00ED (caudatus/putamen). N\u00E1\u0161 pl\u00E1novan\u00FD projekt si tud\u00ED\u017E klade za c\u00EDl zjistit vliv prenat\u00E1ln\u00ED expozice MA na PFC, dorz\u00E1ln\u00ED striatum a dal\u0161\u00ED struktury mesolimbick\u00E9ho syst\u00E9mu. Na z\u00E1klad\u011B na\u0161ich hypot\u00E9z jsme stanovily 7 c\u00EDl\u016F, kter\u00E9 budeme testovat pomoc\u00ED farmakologick\u00FDch, behavior\u00E1ln\u00EDch, elektrofyziologick\u00FDch a molekul\u00E1rn\u00EDch metod. Dopaminergic mesolimbic reward system seems to play a major role in development of drug addiction. Previously we demonstrated that prenatal methamphetamine (MA) exposure impairs postnatal development of rat pups, their cognitive functions in adulthood and affects pain sensitivity and seizure threshold. Our most recent microdialysis work demonstrates that prenatal MA exposure has long-term effects on motor behavior and dopamine metabolism of adult rats. We demonstrated that prenatal MA exposure increases the dopamine level and its metabolites in nucleus accumbens (NAc) after challenge dose of MA (1 mg/kg) in adulthood. It was suggested that a shift of activation prefrontal cortex (PFC) and striatum and there from ventral striatum (NAc) and dorsal striatum (caudate/putamen) is responsible for the change from the first taste of drug and the compulsive behavior (addiction). The present study will, therefore, investigate dopamine system also in PFC, dorsal striatum and other parts of mesolimbic reward circuit. Seven specific aims will test our hypotheses by pharmacological, behavioral, electrophysiological and molecular methods. The results will have an importance in the research of drug abuse and in designing future educational and prevention strategies to minimize these side effects in the progeny."@en . "2"^^ . . .
