In this project we will test our working hypothesis proposing that triggering through different receptors induces engagement of different sets of plasma membrane and cytoplasmic components to produce unique signalosomes around transmembrane adaptor proteins. To this aim we will use strains of mice deficient in the expression of various adaptor proteins, and compare detailed properties of signalosomes in mast cells obtained from normal and mutant mice. New data will be obtained by electron microscopy topographical studies of individual signaling molecules on isolated plasma membrane sheets using conjugates of specific antibodies and colloidal gold.
