"GAP301/11/1730" . "Dietary fiber and polyunsaturated fatty acids(PUFAs), especially omega-3, may function in prevention and therapy of colon carcinogenesis. Based on our previous results we suggest that butyrate and PUFAs alone or in combination induce cellular lipid alterations and influence the effects of cytokines, signal transduction and consequently cell proliferation, differentiation and apoptosis in dependence on cell transformation level, which influence cellular lipidom. After treatment of various colon cell lines with butyrate, PUFAs and their combination we will perform complex analysis of cellular lipid structure and composition and investigate changes of lipid transporter proteins (FAT/CD36, caveolin-1), accumulation of cytoplasmic lipid droplets, oxidative metabolism and events on mitochondria. Next, we will verify the role of fatty acid synthase, activation of signalling pathways (PI3/Akt, p38, ERK1/2, PTEN), and lipid receptor PPARgamma. Finally, we will determine (with therapeutic goal) importance of lipid alterations for cell sensitivity to apoptotic ligand TRAIL."@en .
