The inactivation of specific genes together with point mutation in tumor suppressor Tp53 gene portends poor prognosis and dismal survival. Mutant p53 proteins acquire a -gain of function- activity that mainly contributes to tumor aggressiveness by regulation of a specific set of growth and metastasis promoting genes. In this project we propose to combine molecular biology, bioinformatics and tumor biology to understand, predict and map the role of mutant p53 binding to non-B DNA structures (DNA triplexes) in regulatory regions of mutant p53 target genes (e.g. PPARGC1A, JAK2 and FRMD5). Motivated by our previous research, we will identify new glioma markers on the basis of specific mutant p53-DNA interactions in glioblastoma cell lines using chromatin immunoprecipitation and gene expression analysis. Attention will be aimed to molecular mechanism of how mutant p53 regulates invasive character of gliomas and its possible utilization in tumor treatment by specific drugs."@en .
