"Protein splicing is a unique mechanism of autocatalytic protein editing directly at polypeptide level. Recently, we identified a calcium-dependent self-processing module (SPM) inside the FrpC protein of Neisseria meningitidis that mediates autocatalytic cleavage of the Asp-Pro peptide bond and formation of an isopeptide bond of the newly released carboxyl of the Asp residue with \u03B5-amino group of adjacent Lys residue (protein trans-splicing). The aim of this project is to determine the structure-function relationships underlying the mechanism of SPM-based protein splicing and its potential use in protein engineering. The project will focus on determination of the three-dimensional structure of SPM, which enables a deeper insight into a novel type of autocatalytic cleavage of the Asp-Pro peptide bond and the role of calcium ions in the conformational change of SPM during autocatalytic processing. The capacity of the newly generated fragment to form covalent linkage with amine group will be exploited for development of SPM-based protein ligation technology."@en . "7"^^ . .
