"Nanocrystalline diamond films (NCD) with various physicochemical properties (i.e., roughness and topography, wettability and electrical conductivity achieved by boron doping) will be grown on silicon and titanium substrates by microwave plasma-enhanced chemical vapor deposition. The films will be used as substrates for the adhesion and growth of bone-derived cells and for their phenotypic maturation, manifested particularly by production of collagen, a major protein of the bone extracellular matrix. Collagen will be visualized and quantified by novel optical techniques, namely two-photon excitation microscopy and second harmonic generation imaging. The production of collagen and its maturation into fibrous structures will be modulated by dynamic cultivation of cells in perfusion and rotary bioreactors, and also by electrical stimulation through boron-doped NCD films.
