"In the colon, dietary compounds and endogenous factors regulating cell growth, differentiation and apoptosis operate together, which may have substantial impact determining metabolism and kinetic of colon epithelial cells. Following our previous results, we will investigate the mechanisms responsible for regulation of the dynamics of cell proliferation, differentiation and apoptosis (anoikis) in various colon epithelial cell lines in vitro after their treatment with n-3 or n-6 PUFAs, sodium butyrate, TNF family cytokines and their selected combinations. In this association, changes in cell lipid composition, properties of cell membranes, fatty acid transporters, lipid accumulation, oxidative metabolism and the role of mitochondria, modulation of specific signalling pathways, transcription factors and molecules regulating cell cycle and differentiation-apoptosis transition will be detected. The methods of flow cytometry, fluorescence microscopy, fluorimetry and molecular biology will be mainly used. Our results will bring new knowledge of the interaction of dietary lipid compounds with endogenous regulators in the intestine and enable to deduce the relationships, cross-talk and time sequences of the processes and pathways influencing colon cell kinetics."@en . .
