Better understanding of cytokine responses is conditio sine qua non for the construction of new effective live oral vaccines against Salmonella. Oral vaccines ofSalmonella enterica serotype Typhimurium will be constructed by genetic modification of the virulent LT2 strain. The effect of aromatic pathway gene deletion and stress protein gene deletion on cytokine levels in the small bowel and circulation will be described. The response against oral vaccines will be studied with the aim to show the role of inflammatory cytokines in defence against Salmonella infection and their role in nocious host response and pathogenesis of enteritis. Germ-free and conventional miniature pigs will be used for oral infection with salmonellae, the first ones as microbiologically defined biological model that can be colonized even with non-virulent bacteria.
