"The aim of the proposed project is to work out in vitro methods leading to more effective use of wide spectrum of meiotic and developmental no quite competent bovine oocytes. There will be analysed bovine meiotic incompetent and competent oocytes obtained from follicles 1.5 - 2, 2 - 3 and 3-6 mm in diameter. Oocytes will be tested by two-step culture system, in meiosis inhibiting medium (containing cdc 2 inhibitor - butyrolacton I) and subsequently in meiosis stimulating medium. A biochemical analysis will be performed at different stages of culture. There will be estimated activities of histone HI and MBP kinases, that reflects an p43cdc2 and MAP kinase activities. An immunoblotting with antibodies against p44 (ERK 1 ) and p42 (ERK 2) will be performed to confirm, that the activity of MBP kinase reflected that of MAP kinase.
