Its results should improve our knowledge about some mechanisms of apoptosis and they could be implemented in cancer therapy. The original hypothesis that pro-apoptotic effects of TNF-alpha andanti-Fas could be strengthened by inhibition of the metabolism of arachidonic acid (AA)will be tested, using, among others, non-steroidal anti-inflammatory drugs. Human leukemia HL-60 cells, induced to differentiate by retinoic acid or dimethyl sulphoxide, will be used as a model. The metabolites responsible for modulation of apoptosis will be identified. At the same time, changes of sensitivity of the cells towards apoptotic during the differentiation stimuli will be followed. Changes in the expression pattern of c-Myc, Bcl-2 and Bax proteins will be studied. The experiments will be designed as multifactorial and their results will be analyzed using multiple parameter statistical methods where appropriate."@en . .
