"Tumorigenesis represent a complex, multistage process that may be modulated by genetic polymorphisms in DNA repair and cell cycle controlling genes. Colorectal (CRC) cancer, a common neoplasia, poses serious health problem in the Czech Republic. In sporadic forms low penetrance genes are important for individual susceptibility to the risk of CRC, but high penetrance genes (i.e. tumor suppressor genes, oncogenes and mismatch repair genes) have to be addressed as well. Present project is focused on: A) Molecular epidemiology on DNA samples from CRC patients, aimed at exploring associations between DNA repair polymorphisms, cell cycle and DNA damage recognition genetic polymorphisms and CRC. Novel aspect is the investigation of the role of DNA MMR in the sporadic CRC and simultaneous haplotype analyses of genetic polymorphisms in above genes. B) Molecular genetic part on tissues from colon polyposis with the determination of molecular markers in high penetrance genes relevant in colorectal carcinogenesis. The importance of the Part B is based on the assumption that polyps represent premalignant lesions with the probability of early somatic mutations in critical genes that could turn out to be initiating events and, thus, this will help us to identify the genetic events that lead to the initiation of tumor and its subsequent progression, particularly in sporadic cases, which constitute the largest category of CRC incidence."@en .
