"Coeliac disease (CoD) is a frequent, inflammatory disorder characterised by permanent intolerance to wheat gluten and related prolamins. Gliadin-specific lamina propria CD4+T cells and intraepithelial lymphocytes with cytotoxic activity for gliadin-induced MICA antigens on epithelial cells appear to play fundamental roles in CoD pathogenesis. We participated in the studies suggesting the role of innate immunity cells by demonstrating that\u00A0 mouse macrophages, human monocytic cell lines or dendritic cells can be activated and/or matured by gluten. In this project we shall investigate whether gliadin peptides activate blood monocytes, the mechanism of the activation, and whether the response of monocytes or dendritic cells from CoD patients differ from the response of cells from healthy donors. Furthermore, we shall attempt to identify gliadin peptides triggering activation of macrophages and dendritic cells and to characterize the gliadin receptor molecule. For better understanding of the involvement of gut microbiota in the onset of the disease, we will try to detect and identify bacteria adhering to epithelial surface of jejunal\u00A0mucosa, and analyze\u00A0the composition of bacterial microflora in stool samples of CoD patients and changes associated with gluten-free diet introduction."@en .
