Insuffiency of this inhibitory system represents an important factor in epileptogenesis. The role of this system and especially the synthesis of GABA will be studied in a model of pilocarpine-induced status epilepticus in adult and immature rats. Two types of status epilepticus will be elicited- a nonconvulsive one and a status of severe motor seizures- in rats 12, 25 and 90 days old. An enzyme synthesizing GABA, glutamate decarboxylase, will be studied immunohistochemically(both isoforms, i.e. GAD 65 and GAD 67), and its activity will be measured biochemically. In addition, brain levels of GABA will be estimated. Seizure susceptibility will be tested by means of two convulsants- 3-mercaptopropionic acid (an inhibitor of GAD) and bicuculline (GABA receptor antagonist).
