The project aims at (i.) characterizing structure of the binding sites for endogenously occurring neurosteroid: 20-oxo-5\u00DF-pregnan-3?-yl sulfate (3?5\u00DFS; neusteroid that has an use-dependent inhibitor effext at NMDA receptors), (ii.) elucidate cellular mechanisms involved in the control of synaptic and\u00A0 extrasynaptic NMDA rceptors sensitivity to 20-oxopregn-5-en-3\u00DF-yl sulfate (PS; neusteroid that has a potentiating effect at NMDA receptors) and (iii.) characterize the effects of temperature on the biophysical properties of NMDA receptors and these receptors mediated excitatory synaptic transmission. In this project, recombinant NR1/NR2 receptors expressed in HEK293 cells and synaptically activated NMDA receptors in cultured hippocampal neurons and pyramidal neurons in acutely prepared thin slices from the rat cortex will be used. The aim of the project is to gain new knowledge on the molecular, functional and pharmacological properties of glutamate receptors that is essential in searching for effective approaches to treat some neurological and psychiatric disorders."@en .
