N-methyl-D-aspartate (NMDA), one of prototypic agonists, is able to induce brain damage since the early stages of maturation. The aim of this project is to elaborate and characterizea model neurotoxic damage of immature brain and specify effects of pharmacological blockade of mechanisms involved in NMDA neurotoxicity. Two age groups of rats (7 and 12 days old) will be injected with a small volume of NMDA into one of the following structures: striatum, sensorimotor cortex and hippocampus. Acute effects of NMDA injection will be followed behaviorally and electroencephalographically. Motor development and learning abilities will be tested repeatedly during two months after an insultand then brains will be examined histologicaly (nissl stain) and immunohistochemically (calcium binding proteins, NO synthase) to characterize lost and/or surviving neurons.
