"Right physiological functioning of the intestinal tissue is important for general health of mammalian organism. In colon crypt the epithelial cells migrate upward and on the top die by anoikis associated with cell-extracellular matrix (ECM) interaction loss. The main aim of the project is to demonstrate the role of intestinal epithelial cell anchorage mediated by ECM in maintenance of colonic tissue homeostasis and in colon carcinogenesis. Using various in vitro well-established human colon epithelial cell lines, the changes in focal adhesion complex formation and alteration of associated downstream signal pathways will be identified during different attachment conditions. Moreover, mechanisms of the modulation of cell sensitivity to TNF family members and to differentiation inductor butyrate as the result of the changes in cell-ECM contacts will be studied. Last part of the project will be focused on possible effects of butyrate on TNF-alpha-mediated mestatic potential increase of intestinal epithelial cells. Expected results should contribute to better understanding of pathophysiological behaviour of the intestinal epithelial cells during carcinogenesis and indicating the relevance of the quality and type of anchorage in resulting fate of the cells."@en .
