"Despite great interest in understanding mechanisms regulating self-renewal and onset of differentiation in human embryonic stem (hES) cells, very little knowledge exists regarding growth factor signaling in this particular cell type. It has been demonstrated that hES cells require exogenous basic fibroblast growth factor (FGF-2) to proliferate in an undifferentiated state. Simultaneously, FGF-2 induces ectodermal and mesodermal cells when hES cells are removed from the culture conditions that allow for undifferentiated growth. Therefore, FGF-2 has dual effect on hES cells, which most likely results from differentiation-related changes of FGFR signaling potentials. At the same time, endogenous FGF signaling is believed to play an essential role in the undifferentiated state as well. In this project we will analyze the expression pattern and signaling potentials of FGF-2 and FGFRs in undifferentiated hES cells and hES cells in different stages of differentiation to reveal developmental"@en . . .
