In mice expressing the Ly6.2 halotype, Sca-1 is expressed on many myeloid and lymphoid cells in bone marrow and peripheral blood. Preliminary results showed that stem and progenitor cells are present in Sca-1 negative fraction in C57B1/6 mice. It is desirable to use another surface antigen for isolation of stem cells. We want to separate stem cells on basis of CD117 (c-kit) expression on lineage negative bone marrow cells. We want to characterize these cells by expression of cytokine receptors CXCR-4, c-mpl, anf flt-3. The separated franctions will be tested for hematopoietic reconstitution and also for transdifferentiation into cells of other tissues. We will transplant stem cells from transgenic donor (GFP, ROSA26) into recipients after damage to myocard (doxorubicin), kidney and liver (diclofenac).
