"The rationale of this proposal is to study neurotransmitter release and action at the neuromuscular synapse. New fundamental knowledge is expected to be gained about modulation and regulation of synaptic efficacy. This should help in the search for new physiological and pharmacological tools that could substantially affect the signal transmission in peripheral, and possibly central synapses. We plan to study how the vesicular and non-vesicular acetylcholine (Ach) release is regulated by the nitric oxide pathway and by purinergic agents. Synchronization of quanta release and its modulation by second messenger pathways will also be studied as it can attenuate or strenghten the output postsynaptic potentials.
