Regardless of their localization in various tissues in human organism SC have unique abilities of proliferation, differentiation an plasticity. The aim of proposed project is to characterize three different types of SC isolated from adult tissues (mesenchymal SC from bone marrow, dental pulp SC, and periodontal ligament SC) and compare their resistance to genotoxic stress (ionizing radiation, cytostatics). While hematopoietic SC die after DNA damage by apoptosis, some studies suggest that mesenchymal SC do not react to irradiation by apoptosis, not even after high doses, but only lose their ability to proliferate. This response allows for selection of resistant population, but has a significant downside as it creates a risk of developing a population of cells with damaged genome, if the cells survive in spite of an imperfect DNA repair.
