. "GA303/98/0414" . . . . "The main aim of this project is to introduce human hepatocytes as a model for the study of xenobiotic metabolism and toxicity. Selected natural substances of current and potential use in the prevention and supportive therapy for liver diseases, will be studied using this model. In addition to pharmacologically characterized caffeic acid, rosmarinic acid, quercetin, luteolin, and silybin, already intensivly researched, other substances with prospective cytoprotective effects, baicalin and silybin glycosides, will also be investigated. Model toxins selected will be iron, its chelates with the cytostatic doxorubicin or bleomycin, and tacrine. Primary cultures of rat hepatocytes will serve for comparison. The results will lead to: a) Introduction ofHH model into experimental toxicology in the Czech Republic. b) New information on the toxicity of selected chemotherapeutics. c) Better understanding of the cytoprotective mechanisms of both used and prospective natural substances. d) Characterization o"@en . . . . . .
