"We will study a role of mitochondrial lipoperoxidation & mitochondrial phospolipases A2 in concert with uncoupling due to UCP2 in feedback downregulation of oxidative stress. We will also use UCP2 KO mice. We will estimate a threshold for released free hydroperoxy-fatty acids, above which UCP2 is activated and attenuation of mitochondrial superoxide production exists. Confocal microscopy will be used to study overexpression of UCP2\u00A0 in e.g. HEPG2, INS1-E cells, compared with normal expression or with silenced cells for UCP2 transcript\u00A0 (INS1-E). We will simulate relations between mtPLA2 and UCPn in reconstituted systems and establish whether redox signalling, including signalling by oxidized lipids initiates posttranslational modifications of UCPs, namely whether phosphorylation by oxidative stress-activated mitochondrially located protein kinases affects UCPn uncoupling ability.
