"The aim of the project is to make more accurate (on the basis of genome analysis) diagnostics of staphylococci causing nosocomial infections. By means of quantitative PFGE-macrorestriction analysis, hybridization techniques including ribotyping, PCR, andplasmid analysis, the genomic variability of multiresistant strains of S. aureus as well as strains of the coagulase-negative species, characterized of standard phenotypic tests, will be estimated. Genetic relationship among the strains found by DNA ana lysis will be correlated with the phenytopic properties of the strains. The molecular of DNA hybridization and PCR primers will be prepared from the specific genomic sequences characterizing related groups of organisms. Another research objectives will be to implement pulsed-field electrophoresis in short capillaries into the routine genetic research by combining its enormous separation power with the high sensitivity and selectivity of laser-induced fluorescence detection.
