"Rat mast cells, a widely used model for analysis of anaphylaxis, express on their surfaces large amounts of Thy-1.1 glycoprotein, the aggregation of which results in cell activation. Thy-1 is however not expressed in mouse mast cells indicating differences in cis- or trans- regulatory elements in these two species. To elucidate the role of these elements in species-specific Thy-1 gene expression in mast cells, we will produce transgenic rats carrying genomic clones of mouse Thy-1.2 and will analyze theexpression of the transgenic product in mast cells and other cell types. Thy-1.1/Thy-1.2 rat mast cells would be used for analysis of the role of Thy-1 in mast cell physiology in vivo and in vitro. Alternativly, if cis-regulatory elements play a key ro le in Thy-1.2 gene expression in rat mast cells, we will exploit the structural homology of the rat and mouse Thy-1 gene to map these elements.
