"The project deals with the role of mitochondria during cellular aging and the cross-talk of mitochondrial physiology with the physiology of the aging cell. It will elucidate the signals and molecular mechanisms influencing the role of mitochondria in, and events leading to, aging and apoptosis in yeast mutants in the RAS/cAMP signaling pathway affecting aging, and assess the role of energy metabolism in chronological and replicative aging. The study shall concentrate on the morphology, quantity and respiratory activity of mitochondria and their disruption during aging (with attendant changes in membrane potential, release of cytochrome c and the action of caspases), release of free radicals, oxidative damage of proteins and lipids, cellular antioxidative defense systems and apoptotic markers, as well as the retrograde response (mitochondriato-nucleus communication that leads to an aging-induced change in energy metabolism).
