Dysregulation of gene expression contributes to development of diseases including cancer. One of the most studied regulators of eukaryotic gene transcriptional activation is the protein p53. Our proposal combines the proteomic and genomic approaches in analysis of functional status of p53 and its homologues. We will accompany these approaches with functional and stability analysis of studied proteins, their interactions with different forms of DNA in vitro as well as with analysis of the role of their posttranslational modifications in processes of cell cycle regulation and/or apoptosis. Our project will evaluate effects of inhibiting p53 activation enzymes and p53 homologues on chemo or radiosensitisation.
