"About 300 species of mammals (representing diverse taxa) have adopted a subterranean lifestyle. Their visual systems have undergone extensive convergent regression in adaptation to the lightless underground habitat. Combining various neurobiological and genetic approaches, the proposed project addresses the visual system in strictly subterranean African mole-rats (Bathyergidae). 1) Using opsin-specific markers we shall examine their photoreceptor properties, sequencing of the L- and S-opsin will be used to assess L-opsin gene fate and S-opsin spectral sensitivity. 2) Neurofilament markers, retrograde DiI tracing and intracellular injections of Lucifer Yellow will be used to study the morphology of retinal ganglion cells. The hypothesis that eye regression is coupled with selective elimination of certain ganglion cell types will be tested. 3) Retinal projections will be studied in microphthalmic mammals inhabiting different ecological niches and the prospective presence of niche-specific adaptations will be documented. 4) Retrograde tracing will be used to define the position and size of the visual cortex in adults, and fMRI to examine the timing and dynamics of postnatal sensory reorganization of the occipital cortex. 5) Various markers of apoptosis and neurogenesis in combination with retrograde post mortem tracing will be used to study mechanism of cross-modal sensory compensation."@en .
