"Hemoglobin digestion in ticks is a critical process for tick survival for two main reasons: (1) Blood-meal digestion provides primary energy resources for tick development and egg production; (2) Hemoglobin fragmentation results in formation of peptides with potent antimicrobial activity. Despite its importance, hemoglobin digestion in ticks is almost unknown at molecular level and lags far behind our current knowledge of this process in other hematophagous parasites. Our project is focused on discoveryof the digestive peptidases involved in hemoglobin degradation in the hard tick Ixodes ricinus, the vector of tick-borne encephalitis and Lyme disease. First, we will search for peptidases of cysteine and aspartic class (legumain, cathepsin B, D and L) functioning in tick gut, and determine their cDNA sequences. Second, the identified peptidases will be expressed as recombinant proteins and their biochemical characteristics will be characterized, namely the substrate/inhibitory specificity as well as"@en . . .
