"Aggregation of the high-affinity IgE receptor (FcetaRI) in mast cells and basophils by IgE-antigen complexes results in biochemical events culminating in release of inflammatory mediators. Early events in this process involve translocation of protein tyrosine kinase (PTK) p56/p53lyn and FcetaRI into low-density membrane (LDM) domains and increased tyrosine phosphorylation of several proteins in these domains. Our previous studies suggested that large-size detergent-resistant membrane (DRM) domains, enriched in GPI-anchored proteins and PTK p56/p53lyn are involved in Thy-1-mediated activation of rat basophilic leukemia (RBL) cells. In this proposal we will test our hypothesis that LDM and DRM domains are identical surface structures which are directly involved in the coupling of receptor aggregation to the cell activation.
