"Regulated proteolysis appears to be a central mechanism in controlling gene expression in eukaryotes. It requires cross-talk which seems to be mediated through the homology domains of the main subunits of the PCI complexes (proteasome-COOP9 signalosome-eIF3). In this proposal we suggest to continue the functional analysis of the PCI domain of the subunit eIF3a/Rpg1p of the translation initiation factor 3 of Saccharomyces cerevisiae. Our preliminary results show that this domain is required for stability of eIF3a/Rpg1p, its distribution into stress granules, its interaction with cytoskeleton and reassembly of nocodazole-depolymerized microtubules. We plan to co-localize eIF3a/Rpg1p with markers of various organelles, PCI complexes and RNA metabolism in living cells using intragenous tagging with fluorescent taggs.
