"The main goal of this project is to develop new techniques for pH measurements in liposomes by means of fluorescent probes, including their application to the study of the proton pump of yeast in the proteoliposomes. Steady-state and time-resolved fluorescence studies in both pure and protein-loaded liposomes stained by selected fluorescent dyes will provide better insight into probe response variations due to the changes of the cell medium, occuring when going from buffer solutions to the cell environment. The investigation of liposomal suspensions with pH manipulated using nigericin could help to explain the question why there is a difference in values of pH measured in large cells by fluorescent probes as compared to those obtained directlywith microelectrodes.
