"The main objective of the proposed grant is the research of the changes of the nuclear chromatin structure in the human leukemic myeloid cell lines after the treatment with differentiating agents and after gamma-irradiation. Changes of abl, bcr and c-mycgene topology in cell nuclei will be detected. Translocation t(9,22);(q34,q11) which leads to chimeric bcr-abl gene (Philadelphia chromosome)that plays an important role in the pathogenesis of chronic myelogenous leukemia (CML) will be studied. Fluorescence in situ hybridisation (FISH) combined with the image analysis will be used. Changes in the structure of interphase nuclei will be related to the cell cycle profile and to the stages of cell maturation. In the frame of this project, the reverse transcription polymerase chain reaction (RT-PCR) for c-myc and abl-bcr gene expression will be applied.
