"The aim of present project is to introduce micro-scale isolation procedure as high-throughput one-step purification method for plant tissue samples. The next goal is to develop new class-specific SPE methods based on immunoaffinity chromatography or molecularly imprinted polymers. We propose to use flexible and environmentally friendly SPE sorbents for the highly selective enrichment and efficient clean-up of selected phytohormones from complex plant matrice in a single step prior to their quantitative analysis using hyphenated techniques (Objective 1). In addition, new phytohormone standards and appropriate stable isotope-labelled analogues will be synthesized (Objective 2). We would like to further complete the quantitative measurements by novel chromatographic techniques such as ultra-high performance liquid chromatography or supercritical fluidic chromatography (SFC), and a single sensitive mass spectrometry-based method (Objective 3 & 4).
