Transmission of TBEV is facilitated by tick saliva thanks to modulation of the host immune system. The main aim of the project is to understand how tick saliva and tick cystatins promote viral replication in cells of the innate immune system (dendritic cells, macrophages, and neutrophils). To understand the mechanism of this promotion, IFN-dependent and IFN-independent saliva/tick cystatin effects on TBEV infection will be analysed at the signalling pathway level, and identification of proteins interacting with tick cystatins will be performed. Through the experiments outlined in this proposal we hope to illuminate how the host response to TBEV infection is modulated by saliva and tick cystatins very early after virus entry. Moreover, we expect that the understanding of tick cystatin inhibitory effect on IFN signalling can open the possibility to utilize these tick proteins in the treatment of human immunopathologies where it is desirable to suppress host IFN responses."@en .
