These insults frequently result in long-term consequences later in development up to adulthood. A key role in the pathogenesis of permanent structural and functional deficits is played by complex molecular processes triggered by a massive release of glutamate. Whereas these processes are well described in adulthood, neurodevelopmental aspects have been neglected. We will focus on an assessment of alterations in neurotransmitter levels in the hippocampus and associated functional and morphological changes using models of focal ischemia and glutamate excitotoxicity in immature rats. We will assess the effect of use-dependent NMDA antagonists, with proven neuroprotective effects and minimum of side effects, in models of perinatal brain damage. The understanding of the relationship between the perinatal brain insult, glutamate receptor-mediated excitotoxicity and the consequent morphological changes are very important for developing rational and effective drugs to tread brain insults in children."@en .
