"The exact functions and regulation of Anterior Gradient protein family are poorly understood, but current data identify AGR2 and AGR3 as clinically relevant factors modulating the behavior and response of many cancers in both hormone-dependent and hormone-independent manner. AGR2 predominantly has been shown to be involved in asthma, inflammatory bowel disease, cell transformation, cancer drug resistance, and metastatic growth. AGR proteins belong to the Protein Disulfide Isomerase family with all the key features of endoplasmic reticulum resident proteins. This gives clues into how they may function as oncoproteins through the regulation of protein folding, protein maturation, and protein secretion that can drive metastatic cell growth.
