"This research project aims to characterize specific molecular mechanisms responsible for regulation of light harvesting capacity and dynamics of phycobilisomes and thylakoid membrane proteins in cyanobacteria. We propose to combine methods of molecular biology and modern biophysics, by creating specific mutations proposed to be impaired in state transitions and nonphotochemical quenching (in the French lab) and to analyse them using the SRFI and FRAP methods (in the Czech lab). This project will allow to answer the following points : [1] Role of proteins dynamics during photoprotection [2] Spillover or redistribution of PBs is involved in state transitions? [3] Relationship between the strong orange light induced fluorescence increase and PBs decoupling and its importance in photoprotection [4] Relationship between the strong orange light induced fluorescence increase and state transitions.
